    报告摘要:

    To understand molecular mechanisms of cellular pathways, contemporary workflows typically require multiple techniques to identify proteins, track their localization, and determine their structures in vitro. Here, we developed a visual proteomics approach to address these questions and understand how mammalian sperm are built. Our in situ cryo-electron tomography and subtomogram averaging provided 6.0 Å reconstructions of axonemal microtubule structures. The well-resolved tertiary structures allowed us to computationally match, in an unbiased manner, novel densities with 21,615 AlphaFold2-predicted protein models of the mouse proteome. We identified novel sperm proteins that form an extensive interaction network crosslinking the lumen of microtubules, suggesting roles in modulating the mechanical properties of the microtubule filaments during flagellar beating.

    Dr. Zhen Chen received PhD degree from the Rockefeller University in 2018, and then joined University of California, San Francisco. His areas of research include the in situ structures of human and mouse sperm axonemes by developing cellular cryoET and subtomogram averaging.
